In this study, we investigate the binding interactions between Azithromycin, an antibiotic, and Calf thymusdeoxyribonucleic acid (ct-DNA) as well as human serum albumin (HSA) under simulated physiological conditions at a pH of ۷.۴۰. The examination employs UV absorption spectroscopy and molecular modeling techniques. Our findings regarding Azithromycin's binding to ct-DNA reveal a groove binding mode, with the binding constant computed based on absorption spectral data. Furthermore, UV-vis spectra are utilized to assess conformational changes in the HSA molecule upon the introduction of Azithromycin. Molecular docking analysis highlights the binding of Azithromycin to the major groove of ct-DNA and subdomains IB, IIA, and IIIA of HSA.
