The effects of splenocytes conditioned medium primed with melatonin on the murine breast carcinoma cell line

Introduction: Despite the information available on the anticancer effect and the immunomodulatory effects of melatonin, there is no information on the function of melatonin in the interaction between immune cells and cancer cells. This study aimed to investigate the effects of the conditioned medium of melatonin-treated splenocytes on ۴T۱ breast adenocarcinoma cells. Methods: The isolated splenocytes from Balb/c mice were cultured in the presence of phytohemagglutinin (۱ mg/ml) and different concentrations of melatonin (۰, ۱ /, ۰.۵, and ۱ mM) for ۷۲ hours. The cells were then cultured for ۲۴ hours in the presence of melatonin-free medium and FBS and the conditioned medium was harvested. ۴T۱ cells were exposed to the conditioned medium for ۲۴ h. Results: Melatonin caused a significant increase in the levels of TNF-α, nitric oxide, and myeloperoxidase in the conditioned medium isolated from spleen cells. The viability and proliferative potential of ۴T۱ cells decreased significantly concurrent with regressing the expression of p۵۳ and Bcl-۲ genes and increased expression of caspase-۳ gene after the co-cutler of ۴T۱ cells with the conditioned medium isolated from melatonin-treated splenocytes. Conclusion: Finally, according to the data of this study and the unique role of melatonin in regulating sleep and wake cycles, the importance of proper sleep and receiving natural light in strengthening the immune system becomes more clear.
